Mutation detailsAn IRES-Cre cassette containing a neomycin selection gene flanked with frt sites was introduced into the 3' untranslated region downstream of the translational stop codon and upstream of the endogenous polyadenylation signal. Crossing with a flp deleter line to excise the neomycin resistance gene resulted in animals able to express Cre from the Hoxb1 locus without interfering with endogenous Hoxb1 activity. (J:84448)

Cre expression under the control of the Hoxb1 promoter was used to activate reporter genes in the descendants of rhombomere 4-derived neural crest cells (r4).
  • first expressed at E7 in the caudal primitive streak in mesodermal somites and caudal tail
  • by 10-11 somite stage reporter activity is found in ectodermal cells of r4
  • by E10.5 caudal mesoderm derivatives are found throughout the embryonic trunk and limbs extending rostrally to the caudal hindbrain
  • at E11.5 cells are found in the neuroepithelium and 2nd branchial arch
  • by E14.5, cells are found throughout the embryo
    • in cartilage and bone of the neck and external ear
    • cells of the tongue, salivary gland, pterygopalatine ganglia
    • cell bodies and axon tracts of the facial nerve (VII)
    • CNS localization is primarily to the 7th and 8th sensory ganglion complex and the 7th cranial nerve
