Mutation details: An Igh enhancer and a dominant negative form of the Creb1 gene carrying a serine to alanine subsitution at residue 133 (S133A) was used for the transgene. This mutant form of Creb1 cannot be phosphorylated. The Igh enhancer was used to target transgene expression to lymphocytes. Transgene expression was observed in spleen by Northern blot. A low level of transgene expression was also detected in the thymus. Western blot analysis showed a higher amount of Creb1 protein in resting B cells from transgenic mice compared to wild-type. Less phosphorylated Creb1 protein was also observed in transgenic B cells compared to wild-type.
