Transgene
description
Transgene Type:    Transgenic (Humanized sequence, Inserted expressed sequence)
Mutation:    Insertion

Tg(huS100B)5Daoh expresses 1 gene

Mutation detailsA 17.3 kb fragment of human genomic DNA containing the S100B gene and its transcriptional control elements was used as the transgene. Expression of the human gene in transgenic animals was verified by Northern and Western blot analysis. Strong expression of the transgene was detected in the brain, and very low levels expression was detected in the liver, heart, and kidney. This line carries about 8 copies of the transgene. (J:78970)

Notes
Line 5 was derived from founder H-5, and carries 8 copies of the transgene. Other founders produced were H-2, H-8, and H-4, carrying 110, 70, and 10 copies of the transgene, respectively. The level of transgene expression was found to be directly correlated to the transgene copy number.

Homozygous transgenic mice are viable and fertile. They have impaired learning and electrophysiological disturbances in the hippocampus.
