Mutation details: A mouse Prkcd cDNA tagged with 21 amino acids of bacteriophage T7 major coat protein at the N-terminus, and under the control of the human keratin 14 promoter, was used as the transgene. Human keratin 14 promoter sequences direct expression of the transgene in the basal cells of the epithelium. Western blot also detected expression of the transgene in stomach and thymus of transgenic animals. Expression of the transgene was 8X that of endogenous protein as determined by Western blot analysis. Kinase activity of the transgene product was confirmed by PKC immunocomplex kinase assay.
