Mutation details: A loxP site was inserted into the second intron and a loxP-flanked neomycin selection cassette was inserted into the sixth intron. The neomycin cassette was removed by transient expression of Cre recombinase in ES cells prior to the production of chimeric mice, leaving two loxP sites that flank exons 3-6 in the final allele. Northern blot analysis demonstrated that a transcript of wild-type length is produced from this allele, and western blot analysis confirmed that a normal protein is made.
