Mutation detailsA genomic fragment containing exons 2 and part of 3 was replaced with a lacZ gene with a splice acceptor site and neomycin selection cassette. RT-PCR analysis demonstrated that the mutation eliminated expression of the wild-type transcript, and whole-mount embryo staining studies revealed that beta-galactosidase is expressed from this allele. Homozygous mutant mice display early to mid-gestation lethality (~embryonic day 9.5), random patterning of the left-right body axis, neural tube defects, pericardial sac expansion, enlarged limb buds, and absent embryonic node cilia.
