Abstract:

Full text of MNL contribution: Mouse variants studied by two-dimensional electrophoresis. Several of the previously described mouse variants for soluble liver proteins can also be analyzed using kidney soluble proteins. Proteins found in both tissues, include MUP (LTN-1), LTN-2, LTW-1 (ApoAI lipoprotein), LTW-2, LTW-3, LTW-4, CAR- II (LTW-5), LFO-1 and LFO-2. Liver is preferred for MUP and LTW-4. Kidney is preferred for LTW-3 and CAR-II. Proteins LTH-1 and LTH-2 are not seen in two- dimensional gels of kidney proteins. Four new protein variants have now been found. LTW-6 can be analyzed using liver or kidney proteins. KFO-1, KTH-1 and KTH-2 can be analyzed using kidney proteins. They cannot be seen on gels of liver proteins. LTW-6 has a molecular weight of 21,000. The pI of LTW-6A is 5.0, while the pI of LTW-6B is 5.6. Most strains carry the Ltw-6a allele. Only strains SWR/J, BUB/BnJ, PL/J and MA/myJ carry the Ltw-6b allele. The SWXL RI strains have been scored, but no map position can be assigned at this time. KFO-1 has a molecular weight of 47,000. The pI of KFO-1A is 6.0, while the pI of KFO-1B is 6.1. C57BL/6J and related strains carry the Kfo-1a allele, while DBA/2J, C3H/HeJ and many other strains carry the Kfo-lb allele. The Kfo-1 locus maps very close to Ltw-3 on Chromosome 9. No recombinants have been found among 56 RI strains or over 60 other inbred mouse strains. All tested strains carrying the Ltw-3a allele carry the Kfo-lb allele. KTH-1 is 33,000 molecular weight protein. The pI of KTH-1A is 5.5. The KTH-10 phenotype results in no polypeptide spot at the position of KTH-1 and has been found only in males of a few strains, including AKR/J and C3H/HeJ. RI strain mapping suggests that the Kth-1 locus is on Chromosome 17. KTH-2 is a second 33,000 molecular weight protein. The pI of KTH-2A is 6.8 or higher. The KTH-20 phenotype results in no polypeptide spot at the position of KTH-2. These two Kth-2 alleles are randomly dispersed among the inbred mouse strains. No map position has been assigned.
