  An Flp-dependent alkaline phosphatase reporter locus was generated by inserting a construct containing an frt-flanked PGK-neo gene followed by sequence encoding human ALPP. The inserted construct contained a splice acceptor site at the 5' end. In addition to transcriptional and translational stop signals in the frt-flanked region, the frt-flanked neo cassette was inserted immediately downstream of the ALPP start codon to ensure a lack of ALPP activity in the absence of Flp-mediated excision.
