Mutation details: The transgene consists of a mouse Ret cDNA encoding the RET9 isoform fused to mouse Hoxb7 promoter sequences (-1316 to +81). Mouse Hoxb7 promoter was used to drive expression of the transgene throughout the uretic bud branches. The construct retained all 201 bp of endogenous Ret9 non-coding sequences, including the out-of-frame upstream ATG at position 169, which presumably competes with the bonafide in-frame Ret9 initiation codon at position 201, resulting in a lower level of Ret9 protein. This construct does not generate dominant renal defects.
