Mutation detailsThe transgene was constructed by joining the coding sequence and SV40 polyadenylation signal from the p-EYFP-Tub vector (Clontech), which encodes a fusion protein composed of enhanced yellow fluorescent protein (EYFP) and a human alpha-tubulin, downstream of the chicken beta-actin promoter followed by a Pgk-puromycin resistance cassette. Mitotic spindles of regenerating hepatocytes from partially hepatectomized mice bearing this transgene fluoresce brightly. (J:100110)

Notes
The linearized transgene construct was electroporated into ES cells. Tetraploid aggregation embryos derived from an ES cell clone designated Tub2 were completely fluorescent. Diploid aggregation using this clone generated chimeric mice from which a colony of transgenic mice was derived.
