Notes
  • Experiment
Intraspecific somatic cell hybrids were constructed between mouse lymphoma lines S49.1 and EL4. Cell line S49.1 is glucocorticoid sensitive; cell line EL4 is glucocorticoid resistant. Unselected S49.1 x EL4 hybrids are glucocorticoid sensitive if they retain the complete chromosome complement of the parental lines, including the S49.1 specific glucocorticoid receptor (Nr3c1). Selection of S49.1 x EL4 hybrids for dexamethasone resistance results in specific loss of one Chromosome 18 (181) from the S49.1parent. These results suggest that in S49.1, one Chromosome 18 carries the wild type Nr3c1 gene and the other a mutant Nr3c1 that does not confer glucocortoid sensitivity. Nr3c1 is assigned to Chromosome 18.
