Previously identified anxiety QTL Emo3 was further characterized in this study. Emo3 maps to 73 cM on mouse Chromosome 1.

1.21.2016 Curator Note: Because Emo3 was originally mapped in J:53482 in 1999 using HS stock mice generated from 8 inbred strains, which differ from the population used here,
we consider the current study a separate map experiment and have named the QTL Emo5, emotionality 5.

729 animals from outbred strain MF1 were used to dissect the Emo5 locus by comparing haplotypes to anxiety phenotypes. MF1 is considered by the authors to be a mosaic of 4 different derivative strains: C3H/HeJ, C57BL/6J, AKR/J, and I/LnJ. The Emo5 locus was found to contain 3 separate QTL peaks together accounting for 12% of the variance. Using a technique called quantitative complementation, Rgs2 (78 cM) was identified as candidate gene mapping within the second peak of Emo5. Sequence analysis of the QTL region revealed C57BL/6J as carrying the high allele and either A/J or C3H as carrying the low allele. Supporting the candidacy of Rgs2, this gene is expressed in the brain and mutations in Rgs2 are known to affect behavior.

Rgs18 and B830045N13Rik (Brinp3) map near the 2 other Emo5 peaks and are possible candidate genes for these Emo5 intervals.
