Dyscalc1, a recessively inherited cardiac calcification susceptibility locus, was further examined and fine mapped in several crosses between resistant and susceptible animals. A (C57BL/6J x DBA/2J)F2 intercross mapped Dyscalc1 to 24.5 cM on mouse Chromosome 7 with a maximum LOD score of 9.8 at D7Mit193. (DBA/2J-susceptible; C57BL/6J-resistant) This finding is in agreement with a previous mapping study that mapped Dyscalc1 to 23 cM on chromosome 7 at D7Mit229 using a (C57BL/6J x C3H/HeJ)F2 intercross. (C3H/HeJ-susceptible; C57BL/6J-resistant) Analysis of 12 BXD RI strains also confirmed the position of Dyscalc1 and its mode of inheritance. Finally, recombinant inbred strains derived from C57BL/10 (resistant) donor DNA on a C3H/DiSnA (susceptible) background was used to fine map the Dyscalc1 locus. Strains HcB-12, HcB-24, HcB-28, and an (HcB-24 x C3H/DiSnA)F2 population were screened for chromosome 7 markers spanning the Dyscalc1 locus. Dyscalc1 localized to a 7 cM interval between D7Mit270 (18 cM) and D7Mit82 (25 cM). Potential candidate genes that map to this interval include Hrc, Bax, Myod1, Gprc2a-rs1, and the mouse ortholog of Zin (zinedin).
