Mutation details: ES cells carrying the Hbb-artm1.1Mkg allele, which has most of the Hbb-ar region replaced with a loxP site, were targeted in cis with a construct that replaced the upstream HS-62.5 element with a loxP-flanked neomycin selection cassette. This substitution removed nucleotides -63238 through -59669, relative to the start site of the Hbb-y gene. Transient expression of Cre recombinase eliminated the entire genomic sequence between the newly inserted floxed neo cassette and the loxP site present from the Hbb-artm1.1Mkg allele. This deletion removes genes Olfr64, Olf65, Olf66, and almost all of the Hbb-ar region. (J:139198)

Male ES cells, deltaLCR-D/S-1, were generated from male mice on a 129 background that were heterozygous for the Hbb-artm1.1Mkg allele, carried two different variants of the Hbb genes (HbbD/HbbS) and were hemizygous for the null Hprtb-m3 allele.
