Mutation details: The endogenous locus is interrupted by a construct comprising a loxP site, a neomycin resistance cassette with a strong, triple SV40 polyadenylation signal, and a second loxP site in the same orientation as the first followed by the cDNA sequence encoding firefly luciferase and the bovine growth hormone polyadenylation signal. In cells in which Cre recombinase is expressed, the DNA between the loxP sites is excised, along with one loxP site, permitting expression of the luciferase gene under control of the ubiquitous ROSA26 promoter.
