Mutation details: All of exon 1 and a portion of exon 2 were replaced by a cassette inserted by homologous recombination and containing lacZ and neo. In addition to deleting endogenous sequence, the insertion of the cassette shifted downstream sequence out of frame. Expression analysis of embryonic fibroblasts derived from homozygous mutant mice indicated an absence of both transcript and protein, even after serum stimulation. Expression of lacZ was determined to be driven by the endogenous promoter. (J:80618)
