Mutation details: Insertion of a neomycin resistance cassette into exon 6 immediately 5' of the RGD sequence disrupted the Spp1 gene. Northern analysis of homozygous mutant kidney detected low levels of a high molecular weight RNA species, but no wild-type or truncated Spp1 transcript. Immunoblot analysis detected low levels of a truncated form of the protein in bone but not in other tissues, fluids, or cells of homozygous mutant mice. (J:56535)
