Caption Characterization of the Otoatm1Gpr/Otoatm1Gpr phenotype. Structure of organ of Corti and tectorial membrane (TM) in mature Otoatm1Gpr/Otoa+ (Otoa+/EGFP) (A, C, E, and G) and Otoatm1Gpr/Otoatm1Gpr (OtoaEGFP/EGFP) (B, D, F, and H) mice. (A and B) Toluidine blue-stained, 1-um-thick plastic sections: TM is detached from the spiral limbus (SL) in the homozygous mutant mouse. Arrows indicate marginal band (A) and equivalent region in the mutant (B). (C and D) Transmission EM images showing ultrastructural details of lateral margin of TM. A prominent marginal band (MB) is visible in the heterozygous mutant mouse (C). This region is thinned and fenestrated (arrows) in the homozygous mutant mouse (D). Insets: Details of boxed areas shown at higher magnification reveal imprints of tallest stereocilia of outer hair cells (OHCs). (E and F) SEM of lower surface of TM revealing W-shaped imprints (arrows) of the OHC hair bundles. These are observed around the fenestrae in the homozygous mutant mouse. (G and H) SEM revealing the hair bundles of the inner hair cells (IHC) and three rows of OHCs (01, 02, and 03) in the organ of Corti. The structure and arrangement of these hair bundles is similar in mice of both genotypes. (Scale bars: A and B, 50 um; C-H, 5 um; C and D, Insets, 500 nm).
