Caption Generation of the Dclk2tm1Jgg and Dclk2tm1.2Jgg alleles. (A) Targeting strategy. Exon 2 was targeted by conditional strategy with HSV-TK as a negative and PKG-neo as a positive selection marker. Homologous recombination led to a conditional targeted allele Dclk2tm1Jgg (flox-neo). Complete removal of exon 2 was mediated by Cre recombinase, leading to a germ-line null allele, Dclk2tm1.2Jgg. This allele leads to an out-of-frame mutation and the appearance of a stop codon within the exon 3.
