Caption Generation of the Adam10tm1.1Dhc allele. A targeting vector containing intron 8, exon 9, and intron 9 of mouse Adam10 , and loxP (gray triangles) and FRT (open triangles) sites were inserted into the Adam10 gene via homologous recombination to generate an ADAM10 flox allele. Removal of the Neo cassette via Flp1-mediated recombination generated Adam10tm1.1Dhc (ADAM10delta), containing exon 9 flanked by loxP sites. Cre-mediated recombination can be used to conditionally remove exon 9. Primers P1 and P2 amplify a 955-bp sequence of nonrecombined ADAM10delta or a 217-bp sequence of the recombined allele, respectively.
