The alternatively spliced E12 and E47 exons, respectively, were flanked with loxP sites (indicated by arrows). In addition, a tetracysteine tag (black bar) was inserted into exon 20, 3' of the coding region, to produce tagged E47 and E12 proteins. Furthermore, a neomycin (neo) resistance gene was introduced to screen for targeted ES cells. The EcoRI (E) fragments as well as the probe used for screening of the ES cells are indicated. After crossing of Tcf3tm3Cmu (Tcfe2afl_E12) or Tcf3tm4Cmu (Tcfe2afl_E47)-bearing mice with a germline Cre-deleter strain (Ella-Cre), the corresponding Tcf3tm3.1Cmu (Tcfe2adeltaE12) or Tcf3tm4.1Cmu (Tcfe2adeltaE47) strains were obtained. | ||||||||||
