Caption A, improved survival of Tgfb1tm1(Tgfb3)Kul/Tgfb1tm1(Tgfb3)Kul (LBeta3/LBeta3; mutant) mice (median age = 30.5 weeks) versus the TGF-Beta1-/- mice (median age = 3 weeks) demonstrating rescue from the severe autoimmunity inherent with TGF-Beta1 deficiency. The life span of the mutant mice is still less than the controls, suggesting that TGF-Beta1 and TGF-Beta3 are not completely interchangeable (p =/< 0.0001; log rank (Mantel-Cox) test). B, reduced body weight of male Tgfb1tm1(Tgfb3)Kul/Tgfb1tm1(Tgfb3)Kul mutant mice, which plateaus starting around 5-6 weeks of age. C, photos of female mice showing reduced size and kyphosis in the mutant mice over time. D, faxitron images and photos of a pair of male mice at 27 weeks of age illustrating the hunchback posture of the mutant mouse caused by curvature of the thoracic vertebrae. E, teeth of mutant mice show signs of undermineralization and chipping. The adult mice shown are between 8 and 32 weeks of age. Severe malocclussion was also detected in some of the mutant mice. F, decreased mineral density and dental attrition in the teeth of the mutant mice as shown by faxitron (mice are 15 weeks of age). Using the faxitron images of the teeth, the mineral density of both molars and incisors was measured in the mutant mice (n = 6) and compared with wild-type (WT, +/+) (n = 6). The quantitation in mutant mice incisor opacity revealed a reduction of opacity by about 28% in the upper incisors (Wild-type, 63.7 +/- 14.1 au (arbitrary units); mutant, 45.9 +/- 16 au, and a reduction by 31% in the lower incisors (WT, 111.8 +/- 23 au; mutant, 76.8 +/- 14.5 au (*, p =/< 0.05, mean +/- S.E.)). In the molars, the reduction in opacity was less significant but still present. For the upper molars, there was a 6% reduction in opacity (WT, 126.8 +/- 12.7 au; mutant, 119.2 +/- 19.6 au) and an 18% reduction in the lower molars (WT, 119.2 +/- 2 au; mutant, 97.6 +/- 11.2 au).
