Caption A: Transmission electron microscopy of lung tissue. Images were acquired at high magnification (x33,000). Note that endothelial cells from Cav1tm1Mls/Cav1tm1Mls (KO) mice lack caveolae, whereas their normal counterparts in wild-type (WT) mice showed abundant caveolae (arrows). Endothelial cells were identified by their proximity to red blood cells (RBC) that are electron dense (due to their high iron content) and appear black. Similarly, type I pneumocytes also lacked caveolae in the mutant mice (not shown). Upper panel, wild-type; lower panel, mutant mice. B and C: Immunostaining. Lung paraffin sections from wild-type and mutant mice were immunostained with an endothelial marker, anti-VEGF-R (Flk-1) IgG. Bound primary antibodies were detected with a fluorescently labeled secondary antibody. Arrows point to VEGF-R-positive endothelial cells, which appear more numerous in lung sections from mutant mice. Two representative fields are shown for each genotype. B, wild-type; C, mutant mice. Long overexposures are shown to illustrate better the overall architecture of the adjacent VEGF-R-negative lung parenchyma.
