Mutation detailsIn mice bearing this allele, doxycycline regulates expression of the mCherry fluorescent reporter and three mouse transcription factors (3TF) - v-maf musculoaponeurotic fibrosarcoma oncogene family, protein A (avian) (encoded in mouse by Mafa), pancreatic and duodenal homeobox 1 (Pdx1), and neurogenin 3 (Neurog3) - in cells expressing either the tetracycline transactivator (tTA) or reverse transactivator (rtTA). The allele was generated by recombinase-mediated cassette exchange (RMCE) in ES cells harboring the cassette acceptor allele Gt(ROSA)26Sortm1Mgn. The insertion, flanked by lox71/lox2272 sites, contains a bi-directional tetracycline regulated promoter (a tetracycline responsive element (TRE), comprising seven copies of the tetracycline operator (tetO), flanked by CMV mini promoters) derived from pTRE-Tight-BI driving expression, in one direction, of mCherry, and in the opposite direction, of a single transcript encoding the 3TF separated by 2A peptides, which mediate ribosome skipping via inhibition of peptide bond formation. Excision of a selection cassette has left a single FRT site downstream of mCherry. (J:241149)
