Mutation detailsThe mouse line was generated by homologous recombination using the CRISPR/Cas9 technology. The coding sequences for a viral A2 peptide, followed by the coding sequence for optimized FLP recombinase, was inserted at the termination codon of the endogenous Dbh gene; a single lox2722 (modified LoxP) site marks the former position of a floxed neomycin resistance cassette that was deleted by Cre-mediated recombination. During translation of the bi-cistronic mRNA, the ribosome "skips" the peptide bond between the two C-terminal amino acids (glycine and proline) of the A2 peptide, resulting in production of functional mouse DBH enzyme joined to most of the short A2 peptide and, independently, flpo recombinase with an N-terminal proline.
