Mutation details: An insertion vector composed of a floxed lacZ and polyadenylation signal followed by a GFP tagged human histone H2B and, finally, an FRT flanked neomycin selection cassette was inserted 6 bp upstream of the ATG start site in exon 1. Expression of flp recombinase in properly targeted cells removed the selection cassette. The tagged histone is only expressed after the removal of the floxed lacZ.
