Mutation detailsThe coding sequence of the Sox2 gene in a BAC (RP23-2B8) was replaced with the coding sequence for a tamoxifen-inducible cre/estrogen receptor fusion (cre/Esr1*; CreERTM). The construct was injected into pronuclei of fertilized FVB oocytes. 3 transgene-positive founders were obtained (20, 34, and 36). Founders were bred to reporter mice, administered tamoxifen at E9.5, and embryos were analyzed for recombination activity at E10.5. Line 20 progeny displayed the strongest lacZ expression. This line was characterized further. (J:195589)
