Mutation detailsAn SV40 splice acceptor is fused to a murinized rtTA followed, in order, by the SV40 polyadenylation signal, the 1.2 kb chicken beta-globin insulator, a PGK-hygromycin selection cassette flanked by a loxp and a lox5171 sequence, a promoterless neomycin resistance gene, and, finally, another chicken beta-globin insulator. This insertion is designed so that the floxed hygromycin selection cassette can be replaced by an exchange cassette containing a tetO regulated expression construct and a PGK promoter. Successful exchange places the promoterless neomycin of this insert under the control of the PGK promoter of the exchange cassette. Exchange has been demonstrated to occur as intended in ES cells.
