Mutation detailsTen amino acid substitutions were made in exons 3, 4 and 5 to convert the sequence to a human sequence such that arginine is replaced with a histidine at position 26 (R26H), alanine with glycine at 57 (A57G), arginine with a glycine at 64 (A64G), serine with glutamic acid at 71 (S71E), serine with glycine at 87 (S87G), serine with asparagine at 99 (S99N), arginine with lysine at 100 (R100K), threonine with alanine at 110 (T110A), lysine with arginine at 111 (K111R) and proline to glutamine at 112 (P112Q). A floxed neo cassette used as a selectable marker was removed by expression of cre recombinase. ELISA assays were used to confirm the binding of A4.6.1 to hum-X VEGFA protein expressed from ES cells.
