Mapping and Phenotype information for this QTL, its variants and associated markers

J:40728

Linkage analysis was performed on 520 (CcS-4/Dem x BALB/c)F2 animals to identify QTLs associated with T lymphocyte proliferation in response to IL-2 stimulation. Parental strain CcS-4/Dem is a high responder and parental strain BALB/c is a low responder.CcS-4/Dem is a recombinant congenic strain carrying STS/A-derived donor DNA segments on a BALB/c genetic background. CcS-4/Dem differs from BALB/c at DNA segments on 8 different chromosomes. 23 microsatellite markers covering the differential region weretyped in the F2 animals.

Cinda1 maps to mouse Chromosome 11 near D11Mit4 (37 cM). This locus is linked to T lymphocyte proliferation in response to high levels of IL-2. The BALB/c-derived allele confers increased T lymphocyte proliferation at Cinda1. Potential candidate genes mapping near Cinda1 are Itk (22 cM), Il4 (29 cM), Il5 (29.2 cM), Csf2 (29.5 cM), Il13 (29 cM), Irf1 (29 cM), Ccl11 (47 cM), Ccl7 (46.5 cM), Nos2 (45.6 cM), Ccl1 (47.3 cM), Ccl2 (46.5 cM), Ccl3 (47.6 cM), Ccl4 (47.6 cM), Ccl6 (47.5 cM), Ccl9 (47.4 cM), and Emv14 (37 cM). A previously identified QTL named Tria1 (28 cM) also maps near Cinda1.

Cinda2 maps to mouse Chromosome 12 near D12Mit37 (1 cM). This locus is linked to T lymphocyte proliferation in response to low levels of IL-2. The STS/A-derived allele confers increased T lymphocyte proliferation at Cinda2. Potential candidate genes mapping near Cinda2 are Xmmv39, Mtv30 (1 cM), and Pmv37 (3 cM).
