• cellularity of the thymus is reduced by about 4-fold
• the pre-arranged TCR genes are expressed abnormally early with intracellular TCR-beta chains detectable in the 75% of the common lymphoid progenitors in the bone marrow
• these TCR-beta chains were also detectable in 35% of multipotent progenitors, 12% of short-term hemopoietic stem cells, and 5% of long-term hemopoietic stem cells of the bone marrow
• there is a severe reduction in the percentage of thymocytes at the DN1-3 stages
• however, there is a large population of unusual double-negative thymocytes that are CD3+
• this double-negative CD3+ population is rarely found in the spleen
• there is a greater than 4-fold reduction in the percentage of double-positive thymocytes
• the double-positive thymus makes up 83% of the normal thymus but only 16% in these mutants
• the percentage of CD4+ T cells in the spleen is reduced to less than a third of that in wild-type
• there is a 10-fold increase in the percentage of CD8+ single positive thymocytes
• the percentage of CD8+ T cells among splenocytes is more than double that of controls
• gamma-delta T cells are virtually absent in these mice
