• the CD11b1+ CD2072- CD1032- DC subset is nearly absent in the lymph nodes of immunized mice
• global defect in adaptive immune responses
• when mice are immunized with hapten trinitrophenyl linked to keyhole limpet haemocyanin
• however, no decrease is seen in the T cell independent IgG3 levels
• ovalbumin and aluminum hydroxide or complete Freund's adjuvant with myelin oligodendrocyte glycoprotein peptide induced responses in the lung, lymph nodes and systemic antibody production are significantly reduced
• 18 h after stimulation by fluorescein isothiocyanate skin painting dendritic cells fail to reach the lymph nodes
• this defect can be partially overcome by use of high antigen doses
• splenic DCs fail to relocalize to the T cell zone following LPS stimulation
• however in vitro, bone marrow derived dendritic cells respond normally to a gradient of CCL19 and CCL21, CXCL12, and S1P and no defect is seen in the ability of DCs to transverse an endothelial layer suggesting there is a defect in DC emigration from inflamed tissues
• when injected into wild-type hosts mutant DCs remain near the injection site unlike wild-type cells
• bone marrow derived dendritic cells loaded with antigen are unable to activate T cells
• in lymph node cells from mice sensitized intraperitoneally with OVA/alum and challenged intranasally with OVA
• in lymph node cells from mice sensitized intraperitoneally with OVA/alum and challenged intranasally with OVA
• defect in lipopolysaccharide induced airway inflammation model
