• unlike macrophages from wild-type mice, macrophages from Cias1-deficient mice do not release significant amounts of Il-1beta or Il-18 when primed with LPS and treated with nigericin or maitotoxin to deplete intracellular K+
• null mice secrete much lower serum levels of Il-1beta and Il-18 after receiving a lethal LPS dose
• in response to a lethal dose of LPS, all injected wild-type mice die within 48 hours of caspase-1-dependent endotoxic shock, but only 30% of Cias-null mice die after 72 hours
• macrophages from null mice show reduced Il-1beta and Il-18 release and no caspase-1 activation when infected with S. aureus or Listeria compared to wild-type macrophages in culture
