The authors previously showed that chromosome 8 of A/J mice was associated with susceptibility to S. aureus infection (Ahn, 2010; Yan, 2014). However, the specific genes responsible for this susceptibility are unknown.

Chromosome substitution strain 8 (CSS8) mice, which have chromosome 8 from A/J but an otherwise C57BL/6J genome, were used to identify the genetic determinants of susceptibility to S. aureus on chromosome 8.

Polymorphic microsatellite markers on chromosome 8 between C57BL/6J and A/J were chosen from a database maintained by Mouse Genomic Informatics (MGI). Seventeen microsatellite markers were selected with an average inter-marker distance of 0.65 cM covering chromosome 8. J/qtl software was used to analyze phenotype and genotype data for linkage of survival time after infection with S. aureus.

Phenotypes were defined as either sensitive or resistant based on the dichotomization of survival data (survival of less than 2 day is 0 and survival of longer than 2 days is 1, respectively).

Using 337 S. aureus-infected (C57BL/6J x C57BL/6J-Chr 8A/J/NaJ) mice, QTL mapping identified a region on chromosome 8 that was significantly linked to survival time after S. aureus infection (GRCm38/mm10):

Sstapa4 (suseptibility to Staphylococcus aureus infection 4) maps to Chr 8: 83180780 - 88103009 bp with a peak LOD score of 1.75 at D8Mit80. The A/J derived allele on chromosome 8 that is responsible for S. aureus susceptibility is dominant, and (C57BL/6J-Chr 8A/J/NaJ x C57BL/6J)F1 mice were susceptible to S. aureus.

Cd97 and Crif1 were identified as candidate genes that may play important roles in host defense against S. aureus.
