Mutation detailsA loxP site was inserted upstream of exon 2 and a loxP-flanked MC1-neo cassette was inserted downstream of exon 3. Cre-mediated recombination deleted exons 2-3 and the MC1-neo cassette. Real-time RT-PCR analysis revealed significantly reduced mRNA levels in heterozygous hippocampal dentate gyrus and CA1 regions. Western blot analysis of heterozygous brain tissue confirmed a significant reduction in protein levels, with 73.8% of wild-type levels in the olfactory bulb, 32.1% in anterior cingulate cortex, 37.7% in motor cortex, 30.5% in somatosensory cortex, 44.9% in caudate-putamen (striatum), 43.8% in amygdala, 57.8% in hippocampus, and 47.4% in cerebellum.
