Mutation detailsIn this knock-in line, the coding sequences for nuclear fluorescent protein (Histone H2B-mCherry) and Cre/ERT2 were placed downstream of the Noto coding sequence, with sequences for virus-derived 2A peptides inserted between the protein coding sequences. (2A peptide-containing proteins are self-cleaved downstream of the 2A peptide, such that their introduction allows the production of multiple (3) proteins from a single transcript. The construct also included a neomycin resistance gene in a self-excising ACN cassette (cre recombinase driven by the murine angiotensin-converting enzyme lnked to a floxed neo cassette; the cassette is excised during spermatogenesis). The construct was electroporated into TT2 ES cells. Chimeric founders were crossed to C57BL/6 mice. Deletion of the ACN cassette was confirmed by PCR, (J:153634)
