Mutation
description
Allele Type:    Gene trapped (Reporter)
Mutation:    Insertion of gene trap vector     Vector: pGT0TMpfs     Vector Type: secretory trap

Mutation detailsA gene trap vector randomly integrated in the intron between exons 18 and 19 of the gene. The gene trap cassette contains a splice acceptor that co-opts the endogenous splicing of the mRNA transcript, leading to an in-frame fusion protein containing the first 823 residues of the encoded protein and beta-galactosidase. This gene trap vector also provides a human placental alkaline phosphatase (PLAP) reporter gene expressed through an internal ribosome entry site (IRES). Quantitative PCR analysis using a probe downstream of the gene trap integration site confirmed loss of wild-type transcript in mutant embryos. (J:212179)

Notes
All BayGenomics gene trap mutations were generated in either CGR8 or E14TG2a (129P2/OlaHsd) parental ES cell lines, with the majority in subline E14TG2a.4. The specific ES cell line in which each mutation was made is not specified.
