Mutation detailsThese mice were generated by altering the Tgfb1 codon for C33 to one for S33 by homologous recombination. A floxed neo cassette was included in the targeting vector and was subsequently removed by crossing founder mice with cre transgenic mice. The mutated amino acid is located in the latency-associated peptide portion of the precursor protein that becomes non-covalently associated with the mature but biologically inactive (latent) form of TGF-beta. This mutation disrupts the association of the latency-associated peptide to latent TGF-beta binding proteins. Latent TGF-beta from the sera or platelets of homozygotes is not activated by shear forces. (J:141786, J:142353)
