Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 18, four skeletal trait QTLs mapped to 9 cM (Plskt86; LPR=6.82), 31.7 cM (Plskt87; LPR=5.68), 42.2 cM (Plskt88; LPR=17.34) and 70.7 cM (Scln; LPR=2.32).

Skeletal phenotypes hindlimb obturator foramen length and third caudal length are associated with a locus at 9 cM on chromosome 18 named Plskt86 (pleiotropic skeletal traits 86). The QTL confidence interval spans 6 cM to 15 cM.

Plskt87 (pleiotropic skeletal traits 87) mapped to 31.7 cM and is associated with three skeletal traits in males- face height, interorbital width and mandible square root of angular process area. Plskt87 also displays linkage to body weight at time of necropsy in both sexes. The QTL confidence interval spans 30 cM to 35 cM.

Plskt88 (pleiotropic skeletal traits 88) mapped to 42.2 cM and is associated with seven skeletal traits. The QTL confidence interval spans 39 cM to 47 cM. Skeletal traits linked to Plskt88 include third metatarsal length, first and fourth lumbar lengths, mandible square root of coronoid process area and body weight at 10 weeks of age.

Sacral length in female animals mapped to 70.7 cM and is designated Scln (sacral length). The QTL confidence interval spans 57 cM to 70 cM.
