Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

On mouse Chromosome 10, five skeletal trait QTLs mapped to 12 cM (Plskt49; LPR=7.77, 30 cM (Plskt50; LPR=6.59), 55.6 cM (Plskt51; LPR=9), 70.3 cM (Plskt52; LPR=14.5)and 89.7 cM (Plskt53; LPR=2.55).

Plskt49 (pleiotropic skeletal traits 49) maps to 12 cM and is linked to four skeletal traits- cranial vault length and height, interorbital width and first lumbar centrum height in females. The QTL confidence intervalspans 8 cM to 23 cM.

Plskt50 (pleiotropic skeletal traits 50) is associated with three skeletal traits (mandible square root of condyloid process area, zygomatic length and eighth thoracic length in females). The confidence interval spans 27 cM to 74 cM.

Plskt51 (pleiotropic skeletal traits 51) maps to 55.6 cM and is associated with fourteen skeletal traits. This QTL spans 50 cM to 63 cM. Skeletal traits associated with Plskt51 includes forelimb superior spinous length, skull frontal bone length, ischaic-pubis ramus length, basicranial length, forelimb infraspinous length, face width, mandible square root of angular process area in males, mandible square root of incisor alveolus area, palate length, weight at 10 weeks of age, zygomatic length, necropsy weight and humerus length.

Plskt52 (pleiotropic skeletal traits 52) maps to 70.3 cM and is associated with fourteen skeletal traits including eighth thoracic length, face length, mandible square root of masseteric area and square root of molar alveolar area, mandible square root of coronoid process area, third caudal length in males, zygomatic width, hindlimb ilium length, first lumbar length, hindlimb innominate length, tail length, fourth lumbar length and ulna length. The QTL confidence interval spans68cM to 74 cM.

Plskt53 (pleiotropic skeletal traits 53) is linked to seven skeletal traits at 89.7 cM. The QTL confidence interval spans 85 cM to 90 cM. Associated skeletal traits include first lumbar length, hindlimb ischium length, eighth thoracic length,hindlimb iliac crest length in females, sacral length, first lumbar length and fourth lumbar length.
