Mutation detailsAn 8.2 kb construct containing all the mouse (A/J) H2-Ea kappa exons (from clone lambda Charon 4) plus 2 kb of DNA 5' to the cap site and 1.4 kb 3' to the poly(A) addition site was microinjected into (C57BL/6 x SJL) embryos. Two lines 23 and 16 were generated with line 16 carrying 12 copies of the transgene in its genome and line 23 having 4 copies. Northern blots and S1 nuclease analysis showed transcripts in the spleen. Line 16 produced more transcripts than line 4, and both lines showed correct initiation sites of transcription. (J:111128)

Transgenic lines were derived from two transgenic founder mice and designated line 16 and line 23. Line 16 exhibits slightly higher antibody titers and mRNA levels in response to synthetic antigen.
