origin
Mutant Cell Line:  XB878

Parent Cell Line:  Other (see notes) (ES Cell)
Strain of Origin:  129P2/OlaHsd
Mutation
description
Allele Type:    Gene trapped (Hypomorph, Reporter)
Mutation:    Insertion of gene trap vector     Vector: pGT0pfs     Vector Type: gene trap

Mutation detailsA gene trap vector containing a beta-geo cassette was inserted into intron 4 between exon 4 and exon 5. The insertion creates a 163 KDa fusion protein that contains the N-terminus and acidic domain to the beta-galactosidase and neo coding regions. The presence of the fusion protein was confirmed by western blot analysis. This allele is also a reporter allele. (J:133931)

Notes
All BayGenomics gene trap mutations were generated in either CGR8 or E14TG2a (129P2/OlaHsd) parental ES cell lines, with the majority in subline E14TG2a.4. The specific ES cell line in which each mutation was made is not specified.
Tetraploid complementation analysis of normal appearing embryos at E9.5 and E10.5 reveals grossly normal vasculature and closed neural tubes. Abundant, normal appearing erythrocytes are recovered from E10.5 complementation embryos indicating relatively normal primitive erythropoiesis occurs in absence of Zfp148. No live embryos are observed at E12.5-13.5.
