Mapping and Phenotype information for this QTL, its variants and associated markers

Linkage analysis was performed on 683 animals from a (C57BL/6J x C3H/HeJ)F2 intercross to identify QTLs associated with tibia bone quality traits. A panel of 116 polymorphic markers at an average spacing of 13 cM was used for genome scan. Tibiae from female animals at 4 months of age were subjected to nanoindentation to assess bone hardness and stiffness. C3H/HeJ bones display greater hardness and stiffness compared to C57BL/6J bones.

Significant linkage to tibia hardness (LOD=6) and stiffness (LOD=6.8) mapped to 37.3 cM on mouse Chromosome 8 near D8Mit75. This locus is named Tbqt1 (tibia bone quality traits 1). A previously identified bone QTL named Vtbt5 (38 cM) maps near Tbqt1. A distal locus associated with tibia hardness, named Tbqt2, mapped to 72cM near D8Mit280 (LOD=7.2).

A major locus for tibia hardness (LOD=12) and stiffness (LOD=9.2) mapped to a broad locus on mouse Chromosome 12 between D12Mit153 (15 cM) and D12Mit79 (53 cM). This locus is named Tbqt3 (tibia bone quality traits 3). Peak linkage is near D12Mit214 at 38 cM. Tbqt3 explains 12% of the tibia hardness variance and 10% of the tibia stiffness variance.

Significant linkage to tibia stiffness mapped to 49.7 cM on mouse Chromosome 3 near D3Mit77 (LOD=9.1). This locus explains 3% of the variance and is named Tbqt4 (tibia bone quality traits 4).

Tbqt5 (tibia bone quality traits 5) mapped to mouse Chromosome 13 between D13Mit226 (59 cM) and D13Mit77 (73 cM). This locus is associated with both tibia hardness (LOD=6.9) and tibia stiffness (LOD=6.3) explaining 6% and 5% of the variance, respectively. A previously identified bone QTLs Bmd13 (35 cM) and Vtbt15 (30 cM) map near Tbqt5.

Significant linkage to tibia hardness mapped to 4.1 cM on mouse Chromosome 17 near D17Mit164 (LOD=8.4). This locus explains 3% of the variance and is named Tbqt6 (tibia bone quality traits 6).

Tbqt7 (tibia bone quality traits 7) mapped to 9 cM on mouse Chromosome 19 near D19Mit61 (LOD=6.1). This locus explains 5% of the variance.
