Genetic loci associated with amphetamine-induced locomotor activation were mapped using 34 recombinant congenic strains (RCSs) derived from C57BL/6J and A/J. Eight to 15 animals from each RCS were injected with 5 mg/kg amphetamine solution and measured for total distance traveled in 10 minute intervals. Parental strain C57BL/6J exhibits a greater amount of amphetamine-induced locomotor activation compared to A/J. Six hundred twenty microsatellite markers were used for genetic analysis.

Significant linkage to amphetamine-induced locomotor activation mapped to 12 cM - 13 cM on mouse Chromosome 1 near D1Mit231 and D1Mit372a (P=0.001). This locus is designated Amdt1 (amphetamine distance traveled 1). Homozygosity for C57BL/6J-derived alleles at Amdt1 confers greater locomotor activation.

Highly significant linkage to amphetamine-induced locomotor activation mapped to 12 cM - 19 cM on mouse Chromosome 10 near D10Mit282, D10Mit16, D10Mit106, and D10Mit214. This locus is named Amdt2 (amphetamine distance traveled 2). C57BL/6J-derived alleles at Amdt1 confer increased locomotor activation. Potential candidate genes mapping near this regioninclude Oprm1 (8 cM), Esr1 (12 cM), and Ctgf (17 cM). Amdt2 colocalizes with a morphine preference QTL named Mop2 at 12 cM and overlaps with a fear response QTL, Lrnx8 at 16 cM.

Suggestive linkage to amphetamine-induced locomotor activity was identified at 9 cM on mouse Chromosome 9 near D9Mit90 (P=0.0355) and 50.5 cM on mouse Chromosome 20 near DXMit173 (P=0.0261). *Note-Mapping data in MGI places DXMit173 on mouse Chromosome X.*
