Animal overexpressing the Odc1 transgene Tg(K6ODCtr)55Tgo on a BALB/cJ genetic background are much more resistant to DMBA-induced skin tumors compared to C57BL/6J transgenics. In addition, male BALB/cJ transgenic animals are more susceptible to DMBA-induced skin tumors than female BALB/cJ transgenic animals.

Genetic loci involved in modifying skin tumor susceptibility in transgenic animals were mapped in 136 transgenic (C57BL/6J x BALB/cJ)F1 x C57BL/6J backcross animals. Approximately 100 polymorphic markers were used for the genome scan. At 1 day after birth, animals were treated with DMBA and observed for tumor development up to 22 weeks. Significant linkage was detected at 113.1 Mb on mouse Chromosome 4 near D4Mit52 (54 cM; LOD=3.52). A previously identified adenoma susceptibility QTL named Papg1 (49.6 cM) maps near this locus.

Linkage analysis performed on 263 transgenic (BALB/cJ x C57BL/6J)F2 intercross animals revealed highly significant linkage on mouse Chromosome X (LOD=5.2-6.61) around 89.6 Mb - 92.5 Mb. This locus is named Moo2 (modifier of Odc 2) and peak linkage occurs at DXMit114 (89.6 Mb; 40.2 cM; LOD=6.61). BALB/cJ-derived alleles at Moo2 confer resistance to DMBA-induced skin tumors in Tg(K6ODCtr)55Tgo transgenic animals. Ar (androgen receptor) at 36 cM is a potential candidate gene for Moo2.

In the backcross, suggestive linkage was detected at 24 Mb - 50.9 Mb on mouse Chromosome 17 near D17Mit62 (17.4 cM; LOD=2.34-3.17). In the F2 cross, suggestive linkage was detected at the same location on mouse Chromosome 17 and is thought to be the previously identified Moo1 (modifier of Odc 1) at 23.2 cM.

10.7.2015 Curator Note: Because Moo1 was originally mapped in J:75743 in 2002 using a (C3H/HeJ x C57BL/6J)F1 x C57BL/6J backcross population, which differs from the cross used here, we consider the current study a separate mapping experiment and have named this QTL Moo3. A previously identified UV-induced tumor susceptibility locus named Uvbi2 (89.6 Mb; 23.2 cM) coincides with this locus.

Suggestive linkage to skin tumor susceptibility also mapped to distal mouse Chromosome 6 near D6Mit59 (139.1 Mb; 67 cM; LOD=2.65).
