Mapping and Phenotype information for this QTL, its variants and associated markers

22 chromosome substitution strains (CSSs) were analyzed for 53 complex traits related to sterol levels, diet-induced obesity, anxiety, and amino acid levels. The CSSs are derived from A/J as the donor strain and C57BL/6J as the background strain. The CSS panel consists of 22 different strains, one for each of the 19 autosomes, two for the sex chromosomes, and one for the mitochondria. Donor strain A/J exhibits decreased activity and increased defecation during an open field test compared to background strain C57BL/6J. Donor strain A/J also appears to be more resistant to diet-induced obesity compared to background strain C57BL/6J.

Evidence for 150 QTLs was detected in this study. 8 loci were chosen for mapping analysis to localize the QTLs. F2 intercrosses (or backcrosses) were set up for the individual CSS lines and over 80 animals were genotyped for each cross.

Mouse Chromosome 1 shows linkage to anxiety phenotypes. Linkage analysis mapped a QTL interval between D1Mit151 and D1Mit511. Peak linkage to open field defecation occurs at 95.5 cM (LOD=6.7), peak linkage to total time in light (light/dark test) occurs at 103 cM (LOD=4.1), and peak linkage to first time to exit (light/dark test) occurs at 93 cM (LOD=3.3). This QTL is designated Axtq1 (anxietyQTL 1). Axtq1 maps near a previously identified anxiety QTL named Exq2 (70 cM).

Mouse Chromosome 6 shows linkage to anxiety phenotypes. Linkage analysis mapped a QTL interval between D6Mit138 and D6Mit274. Peak linkage to activity during an open field test occurs at 4.9 cM (LOD=4.1). This QTL is designated Axtq2. Suggestive linkage to open field defecation (LOD=1.6) mapped to 17 cM between D6Mit274 and D6Mit209.

Linkage to diet-induced obesity on a high-fat low-sucrose diet mapped to an interval between D16Mit125 and D16Mit189. Peak linkage occurs at 53.8 cM (LOD=4.3). This locus is designated Diobq (diet-induced obesity QTL). Suggestive linkage to diet-induced obesity mapped to 27.6 cM (LOD=2.7) on mouse Chromosome 6 and to 59.6 cM (LOD=2.3) on mouse Chromosome 10.

Linkage to total time in light (light/dark test) mapped to 51.7 cM on mouse Chromosome 17 between D17Mit39 and D17Mit221 (LOD=3.6). This locus is designated Axtq3.

Suggestive linkage to cholesterol levels mapped to 54 cM on mouse Chromosome 4 between D4Mit31 and D4Mit203 (LOD=2.5).
