Linkage analysis was performed on 167 (C57BL/6 x SPRET/Ei)F1 x C57BL/6 backcross animals to identify QTLs associated with TNF hyporesponsiveness. Parental strain C57BL/6 is sensitive to the toxic effects of TNF whereas parental strain SPRET/Ei and (C57BL/6 x SPRET/Ei)F1 hybrids are resistant. At 8 weeks of age experimental animals were injected with 150 ug of TNF. This dose is lethal to C57BL/6 and 100% lethality is observed 15 to 24 hours after injection. SPRET/Ei and (C57BL/6 x SPRET/Ei)F1 hybrids do not exhibit lethality at this dose. Genome scan was performed using 69 evenly spaced microsatellite markers.

Significant linkage to TNF resistance mapped to 45 cM on mouse Chromosome 6 near D6Mit104 (P=0.00001). This locus is named Tilss1 (TNF-induced lethal shock susceptibility 1). A potential candidate gene for Tilss1 is Tnfrsf1a at 60.55 cM. A suggestive locus mapped to 15 cM on mouse Chromosome 2 near D2Mit417 (P=0.00164). A potential candidate gene for the chromosome 2 locus is Traf2.

Significant linkage to TNF sensitivity mapped to 70 cM on mouse Chromosome 11 near D11Mit333 (P=0.00009). This locus is named Tilss2 (TNF-induced lethal shock susceptibility 2).
