A novel technique was employed to identify QTLs by comparing knockout congenic animals with the background strain. Authors use the B6.129P2-Il10tm1Cgn/J knockout congenic as an example to assess the presence of QTLs affecting open field activity. B6.129P2-Il10tm1Cgn/J displays decreased open field activity compared to background strain C57BL/6J. Authors employ a breeding strategy to elucidate whether the difference is contributed by the knockout mutation or by 129P2 alleles. The breeding strategy canalso elucidate whether the QTL effect is dominant or recessive in nature.

Authors conclude from this experiment that the difference in open field activity between B6.129P2-Il10tm1Cgn/J and C57BL/6J may be attributed to a 129P2-derived gene or genes closely linked to Il10 at 70 cM on mouse Chromosome 1. This locus, named Ofact (open field activity), is recessive. Ofact maps near several previously identified open field activity QTLs on chromosome 1: Opfa (81.6 cM), Axtofa1 (74 cM), and Axtofd1 (74 cM).
