Mapping and Phenotype information for this QTL, its variants and associated markers

J:83832

Linkage analysis was performed on a population of (MRL/MpJ-Tnfrsf6lpr/Tnfrsf6lpr x C3H/HeJ-Tnfrsf6lpr/Tnfrsf6lpr)F2 animals to identify QTLs associated with vasculitis susceptibility. Susceptible parental strain MRL/MpJ-Tnfrsf6lpr/Tnfrsf6lpr develops spontaneous vasculitis whereas parental strain C3H/HeJ-Tnfrsf6lpr/Tnfrsf6lpr is resistant. 103 microsatellite markers at an average spacing of 10 cM was used in the genome scan.

A QTL associated with autoimmune aoritis in MRL mice, Aaom1, was identified at 13.3 cM on mouse Chromosome 4. Peak linkage was detected at D4Mit213 with LOD=3.9. Aaom1 is a male specific locus with the MRL/MpJ-derived allele conferring recessive susceptibility to vasculitis. The QTL interval of Aaom1 is flanked by markers D4Mit266 and D4Mit89. A possible candidate for Aaom1 is the interleukin 11 receptor, encoded by Il11ra1 and Il11ra2. Suggestive linkage to aoritis was also detected at 54 cM on mouse Chromosome 5 with LOD=2.2 at D5Mit23. MRL/MpJ-derived alleles also confer recessive susceptibility to vasculitis at this locus.

Loci associated with vasculitis in the lower limbs were detected on chromosomes 8 and 5. Aevm1 (autoimmune extremity vasculitis in MRL mice 1) mapped to 33 cM on mouse Chromosome 8 with peak linkage at D8Mit31 (LOD=4.5). Aevm1 is a female-specific locus with MRL/MpJ-derived alleles conferring vasculitis susceptibility with additive inheritance. A possible candidate gene for Aevm1 is Il12rb1. Aevm2 mapped to 65 cM on mouse Chromosome 5 with peak linkage at D5Mit136 (LOD=2.3). MRL/MpJ-derived alleles confer dominantly inherited vasculitis susceptibility at this locus.
