Allele Type:    Targeted (Null/knockout)
Mutation:    Insertion

Mutation detailsA neomycin resistance cassette containing stop codons in all three reading frames was inserted into an exon that encodes the extracellular domain. This region corresponds to nucleotides 2962-3370 of the cDNA. Northern blot analyses of intestine from homozygous mutant mice detected two neo fusion transcripts using a cDNA probe for extracellular sequences and did not detect wild-type transcript. A cDNA probe for kinase sequences detected only one of the fusion transcripts. Northern blot analysis of testis dissected from homozygous mutant embryos detected wild-type transcript using a cDNA probe for kinase sequences. Western blot analysis using antibodies against a carboxy-terminal peptide did not detect protein in epididymal extracts from homozygous mutant mice. (J:33259)
